  • Background
    The newly identified Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) poses continual threat to human health due to rapid transmission worldwide. Unprecedented vaccination campaigns have now begun with multiple candidates. To support these studies, ACRO has developed Anti-SARS-CoV-2 subtype antibody Serologic Assay kit including:IgG1, IgG2 IgG3, IgG4.

    The kit is developed for qualitative detection or titer measurement of Anti-SARS-CoV-2 (B.1.617.2) antibody IgG1 (Spike RBD) in human serum.

  • Storage & Shipping
    1. Unopened kit should be stored at 2℃-8℃ upon receiving.

    2. Find the expiration date on the outside packaging and do not use reagents past their expiration date.

    3. The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.

  • Assay Principles
    This assay kit is used to measure the levels of Anti-SARS-CoV-2 antibody IgG1 by employing an indirect ELISA format. The microplate in the kit has been pre-coated with SARS-CoV-2 Spike RBD. First add the diluted samples to the plate and wash the wells after incubation. Afterwards add HRP-Mouse anti-Human IgG1 to the plate and wash the wells after incubation. Lastly the substrate is loaded into the wells and color develops in proportion to the amount of antibody. The reaction is stopped by the addition of a stop solution and the intensity of the color can be measured at 450 nm and 630 nm. The OD Value reflects the amount of antibody bound.

    Your experiment will include 4 simple steps:

    a) Add your sample to the plate, take the Anti-SARS-CoV-2 antibody as Control sample. The samples and Control sample are diluted by Dilution Buffer.

    b) Add diluted Secondary antibody HRP-Mouse-Anti-Human IgG1 to the plate. The Secondary antibody is diluted by Dilution Buffer.

    c) Wash the plate and add TMB or other colorimetric HRP substrate.

    d) Stop the substrate reaction by add diluted acid. Absorbance (OD) is calculate as the absorbance at 450 nm minus the absorbance at 630 nm to remove background prior to statistical analysis. The OD Value reflects the amount of antibody bound.

Detection of Anti-SARS-CoV-2 Antibody, Human IgG1 titer by Indirect-ELISA Assay.
Immobilized SARS-CoV-2 Spike RBD (B.1.617.2) at 1 μg/mL (100 μL/well) can bind Monoclonal Anti-SARS-CoV-2 Antibody, Human IgG1 in 1:50 human serum. Detection was performed using HRP-Conjugated Anti-human IgG1 antibody with sensitivity of 3.9 ug/mL (QC tested).
